Eukaryotic cells are complex systems with multiple compartments. The physical and chemical properties of these compartments largely define biological function, therefore methods that allow us to observe and perturb these properties can help us understand many biological processes. In this lecture, I will give a brief overview of some of the methods that our lab has developed, particularly in the area of small, photoactivatable molecules for single-molecule imaging. Most of the lecture will be devoted to new, yet unpublished, results in the area of protein-small-molecule hybrid probes.
